Chromium speciation is of particular interest due to the differing health effects of its various forms: hexavalent chromium (Cr(VI)) is classified as carcinogenic, while trivalent chromium (Cr(III)) is considered beneficial for human glucose metabolism. The toxicity of Cr(VI) has led to legislation aimed at limiting human exposure. However, analyzing chromium speciation is challenging, as Cr species can easily convert under changing pH and redox conditions, often occurring during sampling or sample extraction.

To prevent human exposure to Cr(VI), various environmental samples are tested for its presence. Industrial waste, such as electronic waste and automobiles, is monitored to prevent environmental emissions. Fertilizers are tested to avoid soil contamination. Aerosols, like welding dust, are analyzed to limit occupational exposure. Products like cement and leather are checked to prevent skin contact with Cr(VI). Drinking water and food are tested to restrict digestive uptake.

Cr(VI) is highly unstable in the presence of organic material, which can act as a reductant, making its presence in such materials transient. Reports of Cr(VI) in items like cereals, bread, milk, body fluids, or animal tissues are often questionable, typically resulting from inappropriate analytical methods (see the news below).

Conversely, Cr(III) is often cited as essential for human nutrition due to its effects on insulin action. However, recent research suggests that chromium may be beneficial only in pharmacological amounts, rather than as an essential mineral (see the news below).

At EVISA, we have never accepted contracts to perform chromium speciation analysis in body fluids or other biological materials because we knew the outcome for Cr(VI) would be none detectable. Additionally, we have critically reported on the acceptance of Cr(III) complexes with enhanced bioavailability in food supplements and vitamin preparations.

Chromium speciation analysis for samples containing organic materials
Chromium speciation analysis is crucial because Cr(VI) is highly toxic and carcinogenic, whereas trivalent chromium (Cr(III)) is generally considered less harmful and, in some cases, beneficial for the glucose metabolism. The toxicity of Cr(VI) is the reason for legislation meant to limit the human exposure. For this reason, different kind of samples from the human environment are analyzed for the presence of Cr(VI). As it is the case with all oxidation species, these species can be easily converted by changing pH and redox conditions during sampling and sample pretreatment. Sample pretreatment is challenging for solid samples, since chromium has to be extracted for most speciation techniques calling for liquid samples.

The Essentiality of Chomium - a Debate
The essentiality of Chromium, particularly in its trivalent form (Cr3+), is praised in most publications dealing with the speciation of chromium. Especially the ambivalence of chromium, with the trivalent form being considered essential, while the hexavalent species is classified as carcinogen, is a very strong argument to differentiate the two species by means of speciation analysis. Yet the essentiality of Cr(III) for human nutrition has been a subject of debate in more recent years.

Yesterday, we welcomed the 22 millionth visitor to the EVISA web portal. Over the past 12 months, more than 2.3 million visitors have used the portal as an information source, viewing an average of 5 pages each.
