The advent of ratiometric fluorescence probes has revolutionised quantitative fluorescence spectroscopy.Using either excitation or emission shifted probes, it is possible to obtain high speed quantitative information regarding the concentrations of various ions in living cells. In microscope-based systems, typically one will use either imaging or photometry, depending upon the fluorescent dye being used and the speed with which one wishes to acquire data.
For high speed work, or for ultra-low light level applications, camera-based systems usually do not have the sensitivity or speed to generate meaningful data. In such cases, a photomultiplier can provide exquisite sensitivity, and accumulate data at very high speeds. This system forms the basis for the RatioMaster™ line of microscopy ratiometric systems.
Single Channel Photometry System This format is used for monitoring excitation-shifted probes such as Fura-2 and BCECF. In this case, a single emission wavelength is monitored by the PMT, while 2 or more excitation wavelengths are toggled by the lamp source. Choose either a standard monochromator, or a high-speed DeltaRam for excitation probes at up to 250 ratios/second. For even faster work, our DeltaScan double monochromator provides dual excitation switching at speeds up to 650 ratios/second. Dual Channel Photometry System Adding a second channel on the microscope photometer allows the simultaneous monitoring of 2 emission wavelengths, useful for such dyes as Indo-1, Snarf and JC-1. Advanced techniques such as FRET (Fluorescence Resonance Energy Transfer) applications are also easily performed. Our FeliX™ software controls all aspects of your data acquisition, and provides acquisition rates up to 1000 points per second. Combined with the exceptional sensitivity of photomuliplier detection, high-speed low-light level experiments are easily performed.
Whichever photometry system you choose, you can easily acquire and analyze data with our FeliX™ software. This powerful paltform will allow you to acquire your data, control shutters, monochromators and other devices, and provide you with a suite of analysis tools to interpret and present your data after the experiment. You can also feed in and acquire as many as 6 additional analog signals SIMULTANEOUSLY with your PMT data. As always, you are free to install this software platform on as many computers as you wish, enabling to analyze and present your data without tying up valuable instrument time.
