The design of the UVD 340U is optimized for maximum sensitivity in the UV range, while maintaining good resolution to distinguish spectra—even of closely related compounds. A high spectrum linearity allows comparison of spectra from trace-level peaks with spectra from higher concentration standards. Discrete, large-area diodes and optimized optics result in an average noise level of less than ±5 µAU (dry cell, 254 nm), providing the low detection limits required for reliable quantification of trace-level compounds. The multidimensional signal extraction option from the 3-D data makes detection wavelength optimization possible in only one chromatographic run. The UVD 340U is operated using the Chromeleon® Chromatography Management System, which includes spectra library screening, peak purity analysis, multidimensional signal extraction, and multiple presentation options for easy method development and data presentation.
The UVD 340U provides the flexibility you need to run all your HPLC applications under optimal conditions. A fused, silica, nano flow cell with 395 nL cell volume and 8-mm path length is ideally suited for applications using 1- to 2-mm i.d. columns (e.g. for microbore LC/MS). For applications using 2- to 3-mm i.d. columns, a micro flow cell with 2.5 µL cell volume and 9-mm path length is recommended. In semipreparative applications, the path length of the flow cell is usually reduced to allow for high sample concentrations. For this purpose, a semipreparative flow cell with 6-µL cell volume and 2-mm path length is available.
USB and Ethernet interfaces make the UVD 340U simple to install and compatible with state-of-the-art communication interfaces. Control through USB provides for plug-and-play installation within minutes. An integrated USB hub allows the connection of up to three additional USB devices directly to the detector. Fast access to the lamp and flow cell minimize instrument downtime during maintenance: the UVD is up running again within minutes. Wavelength verification through a built-in holmium oxide filter ensures correct calibration of the UVD 340U to within ±0.75 mm. Powerful and easy-to-use AutoQ TM validation routines allow for automated Installation Qualification (IQ), Operational Qualification (OQ), and Performance Qualification (PQ) tests. OQ and PQ provide verification of wavelength accuracy, noise, drift, linearity, and lamp intensity.
