Modular Versatility–Integrated Performance
The UltiMate 3000 pumps are supported by a full suite of nano and capillary compatible components. From the in-line, low-volume degassers to the System Wellness monitoring, the UltiMate 3000 exceeds the needs and expectations of nano and capillary LC applications. Ultralow volume flow cells, cooled well plates in a sealed compartment, precise thermostatting of critical flow components, and dual-needle injection principle are just a few of the aspects of the design that make the UltiMate 3000 the best in its class. As a result, the UltiMate 3000 is the perfect companion for mass spectrometry, regardless of the ionization technique (ESI, nanospray, MALDI, etc.).
Nanogradient Accuracy and Reproducibility
Exceedingly reliable generation of nano and capillary gradients is performed using an all-new, groundbreaking integrated active flow splitting system: UltiFlow technology (patent pending). This innovation significantly improves the reproducibility of gradients and flow accuracy compared to passive flow splitting approaches. Presplit gradient formation, with up to three solvents, offers various advantages over splitless nanoflow gradients. Mixing is more homogeneous, gradient delays are minimized, and fast changes to the gradient profiles are optimized even under the lowest column flow rates.
UltiFlow technology also preserves a constant column flow, independent of solvent composition, and column pressures. Even in case of partial column clogging from particulate in a sample, the programmed flow rate will be maintained. UltiFlow constantly monitors flow and immediately initiates corrective action for any deviations from the flow set-point. With the UltiMate 3000 and UltiFlow technology, reliable timing for peaks into the MS or fractions onto a MALDI plate is guaranteed—sample to sample, week to week, and instrument to instrument.
Seamless LC/MS Integration
Nano LC/MS has become an invaluable tool and is the technique of choice in many proteomics applications. Restrictions on sample amounts and the essential demand for increased sensitivity can only be overcome by the use of dedicated nano LC Systems. When applying ESI or nanospray, the separation efficiency, flow accuracy and flow precision are of utmost importance. The UltiMate 3000 exceeds the most demanding criteria for on-line nano LC/MS. A powerful example of the seamless integration is shown in the separation of a cytochrome c digest on a 200 µm i.d. x 5 cm and the corresponding MS and MS/MS spectra obtained by interfacing the UltiMate 3000 to an Ion-trap MS (HCT+, Bruker Daltonics).
For more complex samples, the Flow Manager module with its dedicated microvalves simplifies the setup and execution of column-switching applications. Applications such as 2-D LC (MDLC) are automated and as easy to use as basic single column methods. The performance advantage of the UltiMate 3000 is extended to LC/MALDI with the addition of the Probot™ Microfraction Collector.
The Probot has been specifically designed for highly reproducible, on-line collection of microliter and nanoliter sample with minimal impact on the separation. The figures below demonstrate the MALDI/MS and MS/MS data obtained from a high density MALDI target, analysed using the AB 4700 Proteome Analyzer. An integrated dosage pump adds matrices or make-up solutions as the fraction is spotted onto MALDI targets. Either way, integration is complete and transparent to the performance of the separation.
